Activity-defined at 400 cohesive-end units/µL. 1 cohesive-end unit gives 50% ligation of HindIII-cut lambda DNA in 30 min at 16 °C in a 20 µL reaction. These are cohesive-end units, not Weiss units, so titrate against this definition rather than converting. Applications include sticky-end ligation, blunt-end ligation, nick sealing, Golden Gate assembly, NGS adapter ligation, library construction.
