Nitrilotriacetic acid resin charged with Ni2+ on 6% cross-linked agarose, binding ≥40 mg of 6xHis-tagged protein per mL of resin. The standard IMAC support for His-tag capture under native or denaturing conditions.

Storage buffer30% ethanol
CIPStrip with 100 mM EDTA, wash with 6 M guanidine-HCl or 0.5 M NaOH while uncharged, then recharge with 100 mM NiSO4 and re-equilibrate. Never apply NaOH to a charged bed: the nickel precipitates as hydroxide in the bead and the resin stops binding while still looking blue.
